The win was the second in a row for the Mustangs (5-6), who just three games last season.
"It's extremely hard to go into someone's homecoming," North Pemiscot coach Jamie Johnson said. "I thought Southland played with a lot of emotion tonight."
In other games involving area teams Friday, it was Clarkton 70, Gideon 47; Cooter 65, Delta C7 34; Neelyville 72, Holcomb 61; and Portageville 56, South Pemiscot 43.
The Rebels (1-10), who lost to North Pem 68-49 back in December, led 17-11 after the first quarter and 29-26 at the break before the Mustangs surged ahead.
Southland got 26 points from Justin Kersey and 11 from Jason Cox, but the Mustangs held senior big man Michael George to four points.
"We did a good job on George on the inside and controlling the inside game," Johnson said.
Parker Watkins paced North Pemiscot with 15 points, while Todd Hoggard tossed in 12, Grant St. Cin added 11 and Josh Tomlin finished with 10.
North Pemiscot is back in action Tuesday when it travels to Clarkton, while the Rebels are off until next Friday when they are at Campbell.
Cooter 65, Delta C7 34
At Deering, Devon Barger scored a game-high 18 points for the Wildcats, who steadily pulled away from the host Chargers.
Sloan Richardson scored 16 points to pace Delta (0-7), which hosts Campbell Tuesday while Cooter makes the trip to Risco.
Portageville 56, South Pemiscot 43
At Portageville, the hosts used a 19-12 edge in the third quarter to avenge an upset from last month's Bloomfield Christmas Tournament in a battle of teams called the Bulldogs.
Marcus Greene led Portageville with 16 points while Trey Mosby added 13. Jeremy Patton scored a game-high 20 points to lead South Pem, while teammate Maurichio Clay added 12 in the loss.
